Novel Functions of Death-Associated Protein Kinases through Mitogen-Activated Protein Kinase-Related Signals

Abstract: Death associated protein kinase (DAPK) is a calcium/calmodulin-regulated serine/threonine kinase; its main function is to regulate cell death. DAPK family proteins consist of DAPK1, DAPK2, DAPK3, DAPK-related apoptosis-inducing protein kinases (DRAK)-1 and DRAK-2. In this review, we discuss the roles and regulatory mechanisms of DAPK family members and their relevance to diseases. Furthermore, a special focus is given to several reports describing cross-talks between DAPKs and mitogen-activated protein kinases… Show more

“…Understanding the role of CSCs in BC and their regulatory mechanisms may facilitate therapy and provide a better prognosis. In addition, pathways related to EMT (such as Wnt/β-catenin, Notch, and transforming growth factor-beta (TGF-β) [22]), the sonic hedgehog signaling pathway [69,70], the PI3K/AKT pathway [22], MAPKs [71,72], and the JAK-STAT pathway have been implicated in the development and progression of bladder CSCs [73] (Figure 2).…”

“…However, other novel AD tau associated kinases identified in this study, beyond TBK1 (i.e., DAPK3 and TNIK), also warrant future investigation in their ability to phosphorylate and modify tau. For example, death-associated protein kinase-3 (DAPK3) is a Ca2+/calmodulin (CaM)-regulated serine/threonine kinases with roles in autophagy (64) and TNIK (65) has key roles in dendritic spine structure. Thus, we would hypothesize that tau is a direct substrate for these kinases and has key roles related to the respective functions of DAPK3 and TNIK in brain.…”
