Novel Free-Circulating and Extracellular Vesicle-Derived MiRNAs Dysregulated in Duchenne Muscular Dystrophy

Abstract: Aim: To perform cross-sectional and longitudinal miRNA profiling in plasma from Duchenne muscular dystrophy (DMD) subjects and find non-invasive biomarkers in DMD. Subjects/materials & methods: Plasma was collected from 14 age and sex matched controls and 46 DMD subjects. Free-circulating and extracellular vesicle (EV)-derived miRNA expression was measured by RT-qPCR. Results: Free-circulating and EVs derived miR-29c-3p and miR-133a-3p are dysregulated in DMD subjects. Free-circulating and EV-derived miR-2… Show more

“…But non-coding RNAs in EVs show remarkable stability and more durable activity, and have good RNase degradation resistance. And a higher expression of the EV-derived miRNA pool is likely to result from a shell-like protective activity exerted by the EVs on miRNAs in plasma ( 131 ). More importantly, among the EV bioactive cargoes, non-coding RNAs in EVs are an important component of gene regulation, eliciting either decay or translational repression of target mRNAs while free non-coding RNAs do not have it ( 132 ).…”

“…On the other hand, miR-21-5p and miR-23b-3p decreased in non-ambulant patients with DMD compared with healthy controls [ 93 ]. The same group also assessed both free-circulating miRNAs as well as miRNAs derived from extracellular vesicles (EVs) in serum [ 94 ]. Regarding the free-circulating miRNAs, they found upregulation of miR-1-3p, miR-133a-3p, and miR-29c-3p in patients with DMD compared with healthy controls.…”

“…As stated previously, for now, miRNAs appear to represent the most consequential population of EV cargos. This applies not only to the myomiRs, but also to more ubiquitously expressed miRNAs ( Catapano et al, 2020 ). In animal models of DMD, some miRNAs have been found to be significantly up- or downregulated ( Yedigaryan & Sampaolesi, 2021 ).…”
