Novel Drug Targets and Emerging Pharmacotherapies in Neuropathic Pain

Abstract: Neuropathic pain is a debilitating condition characterized by abnormal signaling within the nervous system, resulting in persistent and often intense sensations of pain. It can arise from various causes, including traumatic nerve injury, neuropathy, and certain diseases. We present an overview of current and emerging pharmacotherapies for neuropathic pain, focusing on novel drug targets and potential therapeutic agents. Current pharmacotherapies, including tricyclic antidepressants, gabapentinoids, and seroton… Show more

“…Moreover, it may have neuroprotective effects, by modulating the activity of various neurotransmitters and receptors involved in pain transmission. NAC also influences excitatory and inhibitory pain signaling in the central nervous system, by interacting with glutamatergic and GABAergic systems and with the activity of ion channels [ 35 ]. Moreover, it may reduce NP, improving the activation of mGlu2/3 receptors and inhibiting the matrix metalloproteinases pathway [ 15 ].…”

“…Findings from numerous preclinical studies indicate the existence of several possible molecular mechanisms operating at the cellular, subcellular, regional, and systemic levels, contributing to LIDO's analgesic properties. These additional mechanisms deepen our understanding of the therapeutic effects of this drug in various pain conditions [22][23][24].…”
