Novel Broccoli-Derived Peptides Hydrolyzed by Trypsin with Dual-Angiotensin I-Converting Enzymes and Dipeptidyl Peptidase-IV-Inhibitory Activities

Abstract: Broccoli-derived peptides show beneficial metabolic effects, and it is necessary to examine their exact functional sequences. First, peptides from the trypsin hydrolysate of broccoli proteins were isolated and identified using column chromatography and quadrupole time-of-flight mass spectrometry. After that, their functions were verified by oral administration. The results identified two novel peptides as Leu-Pro-Gly-Val-Leu-Pro-Val-Ala (LPGVLPVA) and Tyr-Leu-Tyr-Ser-Pro-Ala-Tyr (YLYSPAY). LPGVLPVA exhibited a… Show more

“…The major broccoli proteins were identified, such as jacalin (jacalin-related lectin 34-like isoform X1 and X2), photosystem I, rubisco, ribosomal protein, and other proteins. These identified proteins are different from previous studies ( 20 ).…”

“…It is a pity that there are few studies on the identification of peptides and active peptides of LAB fermented broccoli. Previously, active peptides with hypoglycemic and antihypertensive properties have been identified from broccoli, but peptides with anti-inflammatory properties have not been identified ( 20 ). Meanwhile, the traditional separation and purification of bioactive peptides are very cumbersome and time-consuming ( 21 ).…”

“…OGTT. The oral glucose tolerance test (OGTT) was used to assess the hypoglycemic activity of VPLVM in vivo based on the method by Zhou et al 11 with minor modifications. Healthy C57BL/6 mice (female, 9 weeks old, 21.1 ± 0.4 g) were purchased from Shanghai SLAC Laboratory Animal Co., Ltd. (Shanghai, China).…”

“…8−10 We previously isolated a novel DPP-IV inhibitory peptide VPLVM (IC 50 = 99.68 ± 2 μM) from the protein hydrolysates of broccoli stems and leaves. 11 Peptide VPLVM showed inhibition rates of 88.62% at 0.25 mg/mL and had strong activity versus other food-derived peptides. Nongonierma et al 12 evaluated 25 DPP-IV inhibitory peptides, and the IC 50 ranged from 3.9 ± 1.0 μM (IPI) to 247.0 ± 32.7 μM (FPF).…”
