Novel approaches for the design, delivery and administration of vaccine technologies

Abstract: Summary It is easy to argue that vaccine development represents humankind’s most important and successful endeavour, such is the impact that vaccination has had on human morbidity and mortality over the last 200 years. During this time the original method of Jenner and Pasteur, i.e. that of injecting live‐attenuated or inactivated pathogens, has been developed and supplemented with a wide range of alternative approaches which are now in clinical use or under development. These next‐generation technologies have… Show more

“…Due to safety issues of heat killed or whole cell antigen vaccines, production of plant‐based vaccines has become a striking strategy for the development of cost‐effective vaccines due to some important features such as: high scalability, low health risks, minimum need of cooling chain, and low cost of downstream processing . Currently, delivery of vaccines via plant systems has gained a high consideration platform for oral administration and its renowned potential to stimulate immunity in human and animals .…”

“…The idea of edible vaccine refers to oral administration of antigens expressed in recombinant plants to stimulate the systematic and mucosal immunity against different pathogenic, bacterial, and viral diseases. Upon delivery of vaccine antigen expressed in plants, the cell wall of plants may keep the intracellular substantial and protect them from acidic environment of stomach and within the gastrointestinal tract, these plants would be able to interact with gut‐associated lymphoid tissue (GALT) to stimulate the immune system . These emerging bioencapsulated vaccine antigens expressed in plants provide a cost‐effective platform to deliver different therapeutic proteins in living organisms.…”

“…Vaccination is one of the main strategies to prevent and control infectious diseases [1]. However, during recent decades, many efforts have been being made to obtain alternatives to conventional intramuscular (IM) or subcutaneous (SC) vaccine administration routes, which are mainly administered via hypodermic needles and need trained healthcare personnel [2]. The intradermal (ID) immunization route is of great interest, due to the easy accessibility to the large surface area of the skin and because it has unique immunological properties.…”

“…A lot of research [10][11][12] has been geared toward the understanding of this challenge, but the complexity of it requires a different dimension of approach. 13 An approach that must accommodate many factors affecting vaccine development like pathogen antigenic variability, the emergence of infectious disease, human genetic variation is the goal of immunoinformatics [ Figure 1].…”
