NOTCH1 Aberrations in Chronic Lymphocytic Leukemia

Abstract: Chronic lymphocytic leukemia (CLL) is an incurable B-cell neoplasm characterized by highly variable clinical outcomes. In recent years, genomic and molecular studies revealed a remarkable heterogeneity in CLL, which mirrored the clinical diversity of this disease. These studies profoundly enhanced our understanding of leukemia cell biology and led to the identification of new biomarkers with potential prognostic and therapeutic significance. Accumulating evidence indicates a key role of deregulated NOTCH1 sign… Show more

“…To get a first expression, we filtered for aberrations smaller than 10kb, and for example identified two different SVs affecting NOTCH1 , being a 6kb deletion and a 3.3 kb insertion in sample 14 (CML) and 30 (MDS) respectively. This is a potentially interesting finding, as NOTCH 1 is an important leukemia driver gene in CLL, 34 but also emerges for other leukemia types. 35 Whether these two SVs add anything meaningful to our two cases requires further investigations though.…”

“…There is growing evidence that links CLL to activated NOTCH signaling (7,8). CLL cells have a constitutive NOTCH1 and NOTCH2 activation contributing to apoptosis resistance (9).…”

“…leukemia in adult patients in Western countries [1][2][3]. NOTCH1 mutations associate with clinically aggressive forms of CLL and have emerged as an independent predictor of adverse prognosis [3]. Approximately 80% of NOTCH1 mutations in CLL consist of a 2-bp CT frameshift deletion (c.7541_7542delCT) that generates a truncation in the C-terminal PEST domain and prolongs the half-life of the active form of NOTCH1, i.e., the intracellular domain of NOTCH (ICN) [1,2,[4][5][6].…”
