Notch Signaling Regulation in HCC: From Hepatitis Virus to Non-Coding RNAs

Abstract: The Notch family includes evolutionary conserved genes that encode for single-pass transmembrane receptors involved in stem cell maintenance, development and cell fate determination of many cell lineages. Upon activation by different ligands, and depending on the cell type, Notch signaling plays pleomorphic roles in hepatocellular carcinoma (HCC) affecting neoplastic growth, invasion capability and stem like properties. A specific knowledge of the deregulated expression of each Notch receptor and ligand, coupl… Show more

“…The above-mentioned biological functions and signaling pathways play an important role in promoting tumor development. For example, HIPPO ( 70 ), NOCTH ( 71 ), TGF-β ( 72 ) and WNT ( 73 ) are crucial signaling pathways that regulate various cancer-related processes, including cell proliferation, invasion, metastasis, and immunologic escape. The abnormal activation of these signaling pathways promotes cancer malignancy and leads to a poor prognosis ( 74 77 ).…”

“…Notch signaling pathway regulates hepatocellular carcinoma cell apoptosis by “crosstalk” with other cell signaling pathways or directly affecting internal and external apoptosis pathways ( 38 ). Notch signaling pathway also play an indispensable part in mammalian immunity and cellular homeostasis ( 39 ).…”

“…Abnormalities in Notch signalling were previously described in melanoma, glioblastoma, pancreatic, breast, ovarian cancers and HCC [ 106 ]. Notch signalling is involved in liver progenitor cell differentiation in the setting of cirrhosis with increased inflammation and fibrosis.…”
