Cyanobacteria are well known for their production of a multitude of highly toxic and / or allelopathic compounds. Among the photosynthetic microorganisms, cyanobacteria, belonging to the genus Nostoc are regarded as good candidate for producing biologically active secondary metabolites which are highly toxic to humans and other animals. Since so many reports have been published on the poisoning of different animals from drinking water contaminated with cyanobacteria toxins, it might be assumed that bioactive compounds are found only in aquatic species causes toxicity. However, the discovery of several dead dogs, mice, ducks, and fish around paddy fields, prompted us to study the toxic compounds in a strain of Nostoc which is most abundant in the paddy fields of Iran, using polymerase chain reaction and liquid chromatography coupled with a diode array detector and mass spectrophotometer. Results of molecular analysis demonstrated that the ASN_M strain contains the nosF gene. Also, the result of ion chromatograms and MS 2 fragmentation patterns showed that while there were three different peptidic compound classes (anabaenopeptin, cryptophycin, and nostocyclopeptides), there were no signs of the presence of anatoxin-a, homoanatoxin-a, hassallidin or microcystins. Moreover, a remarkable antifungal activity was identified in the methanolic extracts. Based on the results, this study suggests that three diverse groups of potentially bioactive compounds might account for the death of these animals. This case is the first documented incident of toxicity from aquatic cyanobacteria related intoxication in dogs, mice, and aquatic organisms in Iran.Key Words: cyanobacteria; natural bioactive compound; Nostoc; toxicity INTRODUCTIONThe genus Nostoc is an ecologically, morphologically, and physiologically diverse genus of microorganisms inhabiting soils, and represents large reservoir of potentially valuable natural compounds (Dembitsky and Řezanka 2005). ^Äëíê~Åí= The antifungal compound AK-3 is purified and characterized from intracellular metabolites of the unicellular cyanobacteria, póåÉÅÜçÅóëíáë sp. PCC 6803. AK-3 clearly had antifungal effects upon growth of the fungi ^ëéÉêÖáääìë spp. From 2 g of dry cell powder, 4.8 mg of AK-3 was obtained with a yield of 0.24%. Its structure was elucidated by NMR, UV spectra, amino acid analysis by chemical degradation, and MS measurements, including MALDI-TOF and MS-MS techniques. AK-3 contains a high level of antifungal activity compared to itraconazole and is seemingly a new compound based on its amino acid composition. Extraction was performed with an acetic acid/water mixture (pH 3.0) while detection at UV 214 nm found AK-3 did not react with ninhydrin. A freshwater microalga, Nostoc sp. isolated from Tamdil Wetland of Mizoram was tested for its algicidal, antibacterial and antifungal activity. Extracts were prepared with methanol, ethanol:water and dichloromethane:isopropanol and tested for their antimicrobial activity against nine microorganisms comprising of three chlorophycean algae (Chlorella vulgaris, Scenedesmus quadricauda and Selenastrum capricornatum), one cyanobacterium (Anabaena variabilis), three bacterial strains (Bacillus subtilis, Bacillus pumilus and Escherichia coli) and two fungal strains (Fusarium udum and Fusarium culmorum). Among the three extracts tested the methanol extract was found most effective on algal test organisms. The highest (60%) inhibitory effect was observed against A. variabilis by methanol extract followed by 31% inhibition by dichloromethane:isopropanol. When tested on bacterial strains, the highest inhibition zone (23.67±1.58mm) was observed in B. subtilis by dichloromethane:isopropanol extract followed by ethanol:water extract. In case of fungal activity, the highest inhibition zone (11.00±0.58mm) was observed in F. culmorum by dichloromethane:isopropanol extract and the rest of the extracts showed almost similar effect. MICs of each extract on all the organisms varied. The extracts were further analysed by UV-VIS Spectrophotometer, which reveals the presence of active compounds. Among the three extracts, methanol indicated the highest percentage inhibition and also obtained highest absorption spectra. So, this extract was analysed by GC-MS and we identified the presence of main components in the extract as 1,2-Benzenedicarboxylic acid, mono (2-ethylhexyl) ester, Phytol, n-Hexadecanoic acid, 9,12-Octadecadienoic acid, etc. Further study for purification of the potent compound will explain their usefulness in pharmaceutical and biotechnological industry.
