Norepinephrine: hormone and neurotransmitter in man.

Abstract: To determine whether norepinephrine could subserve a hormonal as well as a neurotransmitter function, norepinephrine was infused for 60 min into each of five normal young men in doses of 0.1, 0.5, 1.0, 2.5, and 5.0 microgram/min. After infusion, the plasma norepinephrine concentration fell with a mean (+/-SD) half-time of 2.4 +/- 0.7 min. The mean (+/-SD) norepinephrine metabolic clearance rate was 3,070 +/- 200 ml/min. The calculated basal plasma norepinephrine production rate was 0.7 microgram/min. The blood… Show more

“…NE not only raises blood pressure but also increases myocardial 0 2 consumption, activates platelets, accelerates ventricular remodeling, induces myocyte apoptosis, and is arrythmogenic. Although the plasma concentrations noted here appear to be below the threshold for any direct cardiovascular or vascular effects, it is the much higher intrasynaptic concentrations associated with such elevations in plasma NE that are cardiotoxic (21).…”

“…21 We have measured the effect of a 12 week course of sibutramine, a serotonin and noradrenaline reuptake inhibitor, on REE and adrenaline induced thermogenesis in 10 obese females and compared the results in 9 obese females receiving a placebo for the same period. Nordrenaline exerts its physiological effects mainly as a neurotransmitter 22 and its metabolic effects as a circulating hormone are only found with exceedingly high plasma concentrations, for example, during prolonged maximal exercise or severe illness like myocardial infarction. 22,23 During hormone infusions, the plasma threshold concentration for cardiovascular and metabolic changes is about 10 times lower for adrenaline than for noradrenaline.…”

“…Results of these studies have largely focused on two particular features: histological evidence of myocarditis (catecholamine cardiomyopathy) 28 " 31 and demonstration of hypertension and bradycardia. 20 -2I -32~34 To determine threshold levels for its metabolic effects, Silverberg et al 35 infused NE in stepwise fashion in doses from 0.1 to 5 ^.g/min over 60 minutes. At the highest dose in these short-term studies (plasma levels of 2150 ± 66 pg/ml), systolic and diastolic arterial blood pressures rose while HR fell.…”
