nor 3′-Demethoxyisoguaiacin from Larrea tridentata Is a Potential Alternative against Multidrug-Resistant Bacteria Associated with Bovine Mastitis

Abstract: Bovine mastitis is one of the most common diseases in dairy cows, and it causes significant economic losses in dairy industries worldwide. Gram-positive and Gram-negative bacteria can cause bovine mastitis, and many of them have developed antimicrobial resistance. There is an urgent need for novel therapeutic options to treat the disease. Larrea tridentata-derived compounds represent an important potential alternative treatment. The aim of the present study was to isolate and characterize antibacterial compoun… Show more

“…The active test compounds with inhibition zones and 5,7-dihidroxyflavanone were further investigated to determine their minimal inhibitory concentration (MIC) using a microdilution method ( Morales-Ubaldo et al, 2022 ). Briefly, in 96-well plates, the stock solutions of the extracts were serially diluted twofold in methanol:water (1:9 v / v ) to final concentrations between 200 to 0.195 mg/mL for extracts, 12.5 to 0.006 mg/mL for C1F1 to C1F10 fractions and 6.14 to 0.006 mg/mL for C2F1 to C2F6.…”

“…The minimum bactericidal concentration (MBC) was performed according to Morales-Ubaldo et al (2022) with slight modifications. Briefly, 10 µL of each sample from the plates where there was no INT color change were added to the agar plates inoculated with a fresh cell suspension (10 8 CFU).…”

“…Morale-Ubaldo et al [ 295 ] isolated and characterized the antibacterial compounds from Larrea tridentate through HPLC techniques against multidrug-resistant bacteria associated with bovine mastitis. The results indicated that nor-3 demethoxy isoguaiacin can be used as an alternative treatment for mastitis.…”

“…It has been reported that carvacrol, hexadecanoic acid, NDGA, and thymol are compounds with bactericidal activity present, as applicable, in the extracts of L. tridentata and O. vulgare obtained with polar organic solvents 3,10,30,31 . It should be noted that, although mechanisms of antibacterial activity of these compounds against most bacteria have not been proposed, the bactericidal capacity has been associated with the alteration of the cytoplasmic membrane 32 .…”
