Nonviral Gene Therapy of the Nervous System: Electroporation

Abstract: Electroporation has been widely used to efficiently transfer foreign genes into the mammalian central nervous system (CNS), and thus plays an important role in gene therapeutic studies on some brain disorders. A lot of work concerning electroporation is focused on gene transfer into rodent brains. This technique involves an injection of nucleic acids into the brain ventricle or specific area and then applying appropriate electrical field to the injected area. Here, we briefly introduced the advantages and the … Show more

“…1 A is adapted from ref. 41 . GOF and LOF studies of Sox9 were performed by coelectroporating PBCAG-mSox9-Flag (1.0 μg/μL) and a CRISPR-Cas9 construct with gRNA against Sox9 exon 1 (1.5 μg/μL, gRNA sequence: 5′- GGTGTTCTCCGTGTCCGAGC-3′), respectively.…”

“…It created temporary pores via electrical pulse in cell membranes through which substances like nucleic acids can pass into cells. Electroporation could almost target all the cells types in vitro , including mammalian cells and bacteria (Ding and Fan, 2016). Electroporation is time saving, labor saving, and cut the cost to a low level.…”
