Nontoxic Intravitreal Dose of Ofloxacin for Rabbit Retina

Abstract: We studied the nontoxic intravitreal concentration of ofloxacin, a new quinolone antibacterial agent, by evaluating its effects on in vitro and in vivo electroretinograms (ERGs) in albino and pigmented rabbits. After perfusion with a 36 μg/ml solution of ofloxacin, the in vitro ERG remained unchanged. The in vivo ERG and the visually evoked potential remained unchanged 4 weeks after vitrectomy with 50 μg/ml ofloxacin, and the retina was within normal limits both ophthalmoscopically and histologically. Therefor… Show more

“…The safety of ocular (topical and/or intra-vitreal) administration of ofloxacin [10][11][12], ciprofloxacin [10], evofloxacin [13], norfloxacin [14], trovafloxacin [15], moxifloxacin [16] and garenoxacin [17] has been investigated, and the results lead to the conclusion that FQs show varied affinity for retinal tissues and there are a marked differences among compounds for safe doses for ocular administration.…”

“…Quinolones have been administered intravitreally in experimental animal models for the possible treatment of endophthalmitis [6,7,16]. This study evaluated the effects of intravitreal trovafloxacin on an experimental rabbit model of S. epidermidis endophthalmitis.…”

“…For instance, the fluoroquinolone antagonism with the GABA receptors was found in the retina, and it was thought to be the explanation of retinal toxicity (Sakai et al, 1994). The FQAs were reported to bind the NMDA receptors in the hippocampal slides (Schmuck et al, 1998).…”
