Nonoxidative ethanol metabolism in humans—from biomarkers to bioactive lipids

Abstract: Ethanol is a widely used psychoactive drug whose chronic abuse is associated with organ dysfunction and disease. Although the prevalent metabolic fate of ethanol in the human body is oxidation a smaller fraction undergoes nonoxidative metabolism yielding ethyl glucuronide, ethyl sulfate, phosphatidylethanol and fatty acid ethyl esters. Nonoxidative ethanol metabolites persist in tissues and body fluids for much longer than ethanol itself and represent biomarkers for the assessment of ethanol intake in clinical… Show more

“…Of ethanol’s metabolites, acetaldehyde is widely considered to be the most potent in colorectal carcinogenesis through its interactions with various cellular and biochemical processes [ 10 , 12 , 51 , 53 , 54 , 55 ]. Non-oxidative metabolism, although occurring at lower rates, has a range of physiological influence that can be quite profound due to the wide variety of metabolites produced [ 11 , 56 ]. This variety is derived from the ability of ethanol to “insert” itself into the structures of significant biochemical molecules [ 56 ].…”

This variety is derived from the ability of ethanol to “insert” itself into the structures of significant biochemical molecules [ 56 ]. For example, ethanol can undergo an enzymatic esterification with fatty acids (FA) to produce fatty acid ethyl esters (FAEE) via FAEE synthase (FAEES) [ 56 ].…”
