Nonhuman primate models of NeuroAIDS

Abstract: Human Immunodeficiency virus (HIV), the virus that causes acquired immunodeficiency syndrome (AIDS), also manifests neurological complications. HIV-associated dementia (HAD) is the most severe form of HIV-induced neurocognitive disorders. HIV encephalitis (HIVE), the pathological correlate of HAD, is characterized by the formation of multinucleated giant cells and microglial nodules, astrocytosis, and neuronal damage and loss. Pathological evaluation of HAD disease progression in humans is not possible, with t… Show more

“…Infection of rhesus macaques with pathogenic simian immunodeficiency virus (SIV) clones and/or viral swarms is a widely used model for HIV/AIDS (13) as well as neuroAIDS (14). SIV-infected macaques exhibit clinical manifestations similar to those of HIV-1-infected humans, albeit on a shorter time scale of approximately 1 to 3 years (13), and the incidence (Ͻ25%) of SIV-associated encephalitis (SIVE), the pathological hallmark of neuroAIDS, is similar to that of untreated seropositive patients (Ͻ30%) (15).…”

“…Although combination antiretroviral therapy (cART) has markedly diminished the incidence of overt HAD, a milder-to-moderate form of neurocognitive impairment known collectively as HIV-1-associated neurocognitive disorders (HAND) developed in an estimated 50% of treated patients (1)(2)(3). Lentiviral neuroinvasion involves the recruitment of infected monocytes/macrophages and/or T cells to the central nervous system (CNS) from circulation, with activation and productive infection of resident microglia and a more restricted/latent infection of astrocytes (4)(5)(6)(7)(8)(9). HIV-1 infection of mononuclear phagocytes in the CNS has been suggested to be the critical driving force of HIV-induced encephalitis (HIVE), as well as HAND (10,11).…”

“…15,20 MRI scans were performed on a Siemens 3T Trio scanner with the CP extremity coil (Siemens Medical Solutions, Malvern, PA) before SIV inoculation and in 2,4,8,12,16, and 20 weeks post-inoculation (WPI). Blood samples were collected from each animal for monitoring CD4 + and CD8 + T cells before each MRI scan.…”

“…The simian immunodeficiency virus (SIV)-infected macaque models exhibit neuropathological symptoms similar to those seen in HIV-infected humans, and have been widely used for studying the cognitive and neuropathological sequelae of AIDS [14][15][16][17] or vaccine development. 18,19 SIVsmmFGb is a novel simian lentivirus strain inducing neuropathology in over 90% of infected pig-tailed macaques.…”
