Nonarteritic ischaemic optic neuropathy (NAION) after 36 h of intake of sildenafil citrate: first Egyptian case

Abstract: Sildenafil citrate is a selective phosphodiesterase 5 (PDE-5) inhibitor and partial phosphodiesterase 6 inhibitor prescribed for erectile dysfunction. Post-marketing case reports of nonarteritic anterior ischaemic optic neuropathy (NAION) over the past few years suggest a potential link with PDE-5 inhibitors. We report a case of a 48-year-old male patient who had acute vision loss 36 h after the intake of 50 mg sildenafil citrate. NAION occurred at a period of minimal blood level of sildenafil citrate. So, ere… Show more

“…Since 2001, sporadic cases of NAION due to sildenafil use have been reported [3, 4, 5]. In all these cases, the affection was unilateral and most had a small, crowded optic nerve head (‘disc at risk’).…”

“…Increased cGMP results in relaxation and vasodilation of vascular smooth muscle [1, 2] and enhances penile erectile function in men. Some reports suggest that sildenafil use may be associated with nonarteritic anterior ischemic optic neuropathy (NAION) [3, 4], and several patients have developed NAION between 1 and 12 h after taking sildenafil [5]. We report, for the first time, to our knowledge, a case of bilateral simultaneous anterior NAION after taking sildenafil.…”
