Non-Steroidal Steroid Receptor Modulators

Abstract: The last ten years much attention has been focused on the finding of non-steroidal ligands for steroidal nuclear receptors for reasons such as diminishing cross-reactivity to eliminate side effect profiles, changing physicochemical properties which might cause different tissue distribution profiles and altering binding modes which influence the binding of cofactors. Compounds with a selective functionality profile are referred to as selective nuclear receptor modulators (e.g., SARMs or SPRMs). In the following… Show more

“…Similarly, and despite the fact that androgens have beneficial effects on bone and muscle mass, their negative impact on the prostate prevents their widespread use in the treatment of osteoporosis and frailty in males (20). This dual and important medical need has led to the development of several SERMs and the marketing of some (1,5,8) as well as the search for selective androgen receptor modulators (SARMs), some of which are currently in preclinical or clinical development (21,22).…”

“…The latest developments in nonsteroidal AR ligands have been extensively reviewed in last five years, including the structural biology and structure-activity relationships (1), medicinal chemistry (2), and potential clinical applications (3). This review focuses on the in vivo pharmacokinetic and pharmacologic properties of nonsteroidal AR ligands, including antagonists, agonists, and selective androgen receptor modulators (SARMs).…”

“…Recent peer-reviewed and patent publications have disclosed SARMs with improved pharmacokinetic properties and tissue selective activity. A handful of excellent reviews on non-steroidal AR ligands have been published [2][3][4][5][6][7][8][9]. This review will provide a comprehensive overview of the SARM literature up to August 2005 and will put particular emphasis on the therapeutic potential of this exciting class of molecules.…”
