BACKGROUND AND AIM:Increasing incidence of medically compromised patients seeking dental treatment and implant rehabilitation, necessitates greater knowledge toward managing such patients. The objective of the present study was to evaluate the perceptions of dental interns in Riyadh, Saudi Arabia, toward dental implant therapy for medically compromised patients.MATERIALS AND METHODS:Using a convenience random sampling technique, a cross-sectional questionnaire-based study was conducted to evaluate the perceptions of the interns from dental schools in Riyadh, Saudi Arabia. Data pertaining to demographic details of the participants, academic grade point average (GPA), clinical implant training and experience and perceptions about dental implant therapy for medically compromised patients were collected.RESULTS:The survey response rate was 82.9% (n = 174/210; Males-129/Females-45). Less than half of the interns (n = 82/174; 47.1%) had performed dental implants, out of which 41.5% (n = 34/82) had placed implants in medically compromised patients. Most medical illnesses except controlled diabetes mellitus and hypertension were perceived by the interns as contraindications for dental implant placement.CONCLUSION:Based on the results of the present study, it could be concluded that courses with a greater emphasis toward management of dental patients with medical problems and undergraduate implant training for such patients would result in better knowledge and perception among dental interns regarding dental implant placement in medically compromised patients.

Among hundreds of thousands of signal receptors contributing to oncogenic activation, tumorigenesis, and metastasis, the hepatocyte growth factor (HGF) receptor-also called tyrosine kinase MET-is a promising target in cancer therapy as its axis is involved in several different cancer types. It is also associated with poor outcomes and is involved in the development of therapeutic resistance. Several HGF/MET-neutralizing antibodies and MET kinase-specific small molecule inhibitors have been developed, resulting in some context-dependent progress in multiple cancer treatments. Nevertheless, the concomitant therapeutic resistance largely inhibits the translation of such targeted drug candidates into clinical application. Until now, numerous studies have been performed to understand the molecular, cellular, and upstream mechanisms that regulate HGF/METtargeted drug resistance, further explore novel strategies to reduce the occurrence of resistance, and improve therapeutic efficacy after resistance. Intriguingly, emerging evidence has revealed that, in addition to its conventional function as an oncogene, the HGF/MET axis stands at the crossroads of tumor autophagy, immunity, and microenvironment. Based on current progress, this review summarizes the current challenges and simultaneously proposes future opportunities for HGF/MET targeting for therapeutic cancer interventions.

Autoimmune diseases such as multiple sclerosis (MS), type I diabetes (T1D), inflammatory bowel diseases (IBD), and rheumatoid arthritis (RA) are chronic, incurable, incapacitating and at times even lethal conditions. Worldwide, millions of people are affected, predominantly women, and their number is steadily increasing. Currently, autoimmune patients require lifelong immunosuppressive therapy, often accompanied by severe adverse side effects and risks. Targeting the fundamental cause of autoimmunity, which is the loss of tolerance to self-or innocuous antigens, may be achieved via various mechanisms. Recently, tolerance-inducing cellular therapies, such as tolerogenic dendritic cells (tolDCs) and regulatory T cells (Tregs), have gained considerable interest. Their safety has already been evaluated in patients with MS, arthritis, T1D, and Crohn's disease, and clinical trials are underway to confirm their safety and therapeutic potential. Cell-based therapies are inevitably expensive and time-consuming, requiring laborious ex vivo manufacturing. Therefore, direct in vivo targeting of tolerogenic cell types offers an attractive alternative, and several strategies are being explored. Type I IFN was the first disease-modifying therapy approved for MS patients, and approaches to endogenously induce IFN in autoimmune diseases are being pursued vigorously. We here review and discuss tolerogenic cellular therapies and targeted in vivo tolerance approaches and propose a novel strategy for cell-specific delivery of type I IFN signaling to a cell type of choice.
