Nomograms Predict Survival Advantages of Gleason Score 3+4 Over 4+3 for Prostate Cancer: A SEER-Based Study

Abstract: Background: Different proportions of Gleason pattern 3 and Gleason pattern 4 lead to various prognosis of prostate cancer with Gleason score 7. The objective of this study was to compare the survival outcomes of Gleason score 3+4 and 4+3 based on data from the Surveillance, Epidemiology, and End Results cancer registry database, and to investigate independent prognosis-associated factors and develop nomograms for predicting survival in Gleason score 7 prostate cancer patients. “…Among the diagnostically significant parameters, sPSA and GS are well-established independent predictors of high-grade PCa, whereas family history is an independent predictor of treatment outcome. 24 , 25 In this study, none of these covariates was found to be significantly associated with MAOA except sPSA. Because high sPSA is known to be a useful predictor of bone scan-positive metastasis in PCa patients, this finding further favors association of MAOA with skeletal metastasis.…”

“…The predictive capacity of models was summarized using timedependent ROC curves [12,13]. The calibration refers to the closeness between the predicted probabilities and the actual outcomes, and this was validated using calibration plots [14]. To test the clinical value of the predictive new model, we generated a decision curve analysis graph to visualize the potential net benefit between two models at each threshold probability [15,16].…”

“…The bioinformatics method based on the TCGA database has been increasingly used to analyze the molecular basis of prostate cancer development and clinical patient prognosis [14][15][16]. By using appropriate analytical software and methods to explore a variety of large-size data of clinical specimen from the TCGA database (including transcriptome sequencing data, gene sequencing data, ATAC-seq data, etc.…”
