Nodal Signaling in Early Vertebrate Embryos

Abstract: The nodal family of TGFbeta-related ligands have emerged as critical regulators of early vertebrate embryogenesis. Recent studies in mice, fish, and frogs of nodals and their intracellular transducers allow a comparison of how this signaling pathway is used in the patterning of early embryos of these different vertebrates.

“…Briefly, Nodal ligands are secreted molecules that bind the Activin receptors ActRIB (ALK4), ActRIIA, ActRIIB, and possibly ALK7 (Kumar, 2001;reviewed in Schier and Shen, 1999;Whitman, 2001;Schier, 2003). This binding appears to necessitate EGF-CFC coreceptors: Cripto and Cryptic in mammals, oep (one-eyed pinhead) in zebrafish, FLR-1 in frogs, and CFC in chicks.…”

“…One of the first steps was the cloning of "activin receptors"-receptors that certainly bind and allow activin signaling but may have a broader range of TGF-␤ superfamily ligands. These were found to be transmembrane serine-threonine kinases and, like TGF-␤ receptors, function as het-erodimers of a Type I and Type II receptor protein (reviewed in Whitman, 2001;Attisano and Wrana, 2002). Kinase-dead mutant versions were made and used as dominant negative reagents.…”

“…Mouse and zebrafish genetics have given the best evidence for the existence in vivo of an essential dorsalspecific activin-like activity, and these genes have been identified as the nodal gene in mouse and the nodalrelated genes cyclops and squint in zebrafish (reviewed by Whitman, 2001). The nodal proteins are, like activin, members of the TGF-␤ superfamily.…”

“…The Nodal ligand binds a receptor complex that includes Activin type 1 and type 2 receptors (Whitman, 2001), together with an EGF-CFC-type coreceptor (Shen et al, 1997;Ding et al, 1998;Zhang et al, 1998;Dorey and Hill, 2006;Onuma et al, 2006). Lefty, a divergent member of the TGF␤ family whose transcription is directly regulated by Nodal signaling, is one of the principal extracellular feedback inhibitors of Nodal signaling (Meno et al, 1996(Meno et al, , 1999Thisse and Thisse, 1999;Cheng et al, 2000;Tanegashima et al, 2000;Branford and Yost, 2002;Cha et al, 2006).…”
