Nociceptive tolerance is improved by bradykinin receptor B1 antagonism and joint morphology is protected by both endothelin type A and bradykinin receptor B1 antagonism in a surgical model of osteoarthritis

Abstract: IntroductionEndothelin-1, a vasoconstrictor peptide, influences cartilage metabolism mainly via endothelin receptor type A (ETA). Along with the inflammatory nonapeptide vasodilator bradykinin (BK), which acts via bradykinin receptor B1 (BKB1) in chronic inflammatory conditions, these vasoactive factors potentiate joint pain and inflammation. We describe a preclinical study of the efficacy of treatment of surgically induced osteoarthritis with ETA and/or BKB1 specific peptide antagonists. We hypothesize that a… Show more

“…These results are relevant because pain and oedema are hallmark responses of an acute gouty attack in humans 8 9 37. Similar to our findings in a gout model, IA administration of a B 1 R antagonist is able to reduce pain and joint damage in a model of osteoarthritis 38. Additionally, since IA administration is routinely used clinically, it is possible that local administration of B 1 R antagonists could be an effective way to treat gouty attacks in humans 8…”

“…The genesis and progression of OA is closely associated with cartilage injuries as a result of arthritis (19). When cells are under inflammatory stress or injured, abnormal mechanical stress results in activation of cartilage cells to stimulate macrophages to produce a series of inflammatory mediators.…”

“…The exponential decay of average ROI image signal intensity (SI) with increasing echo time (TE) was plotted and a least-squares best fit was applied. The best fit decay curve was modeled with the equation: SI (TE) = M 0 −(TE/T2) , allowing for the calculation of the spin density (M 0 = SI (0)) and T2 relaxation time (T2) for the given ROI 45, 46 .…”
