Nocardia asteroides strain GUH-2 induces proteasome inhibition and apoptotic death of cultured cells

Abstract: Many bacterial pathogens have the ability to induce apoptosis in their hosts. It was previously shown that Nocardia asteroides strain GUH-2, a Gram-positive facultatively intracellular pathogen, is capable of inducing the apoptotic death of dopaminergic cells in the murine brain and in PC12 cells, a rat cell line. In this study, the apoptosis-inducing potential of N. asteroides GUH-2 was further explored using HeLa cells, a human epithelial cell line. HeLa cells were incubated for 5 hours with live nocardiae, … Show more

“…One possible mechanism could involve lysosomal acid phosphatase, which has been shown to correlate with intracellular degradation of N. farcinica by murine macrophages (12). At MOIs of Ն1, N. farcinica induced apoptosis of human DCs, and similar data have been reported for the effect of N. asteroides on human epithelial cells (6).…”

“…In Hela and PC12 cells, live Nocardia promoted a significant DNA fragmentation during 6 h-exposure period (Tam et al, 2002; Barry and Beaman, 2007). Previous studies also showed that Nocardia localized in substantia nigra and then underwent a rapid growth, inducing apoptosis of dopaminergic cells and failure of inflammatory response in 24 h (Kohbata and Beaman, 1991; Tam et al, 2002).…”

“…How could GUH-2 induce a cortical Lewy body? Barry and Beaman demonstrated that log-phase GUH-2 secreted a proteasome inhibitor [12]. This inhibitor appeared to be involved in the induction of cortical Lewy bodies, as well as the apoptotic death of dopaminergic cells [10,12,18].…”
