“…Most of the published results on this topic have been obtained in ovariectomized rats treated up to 3 months with C. racemosa extracts [11,23]. The immature uterotrophic assay we used, more sensitive than the ovariectomized uterotrophic assay [24], confirmed that with black cohosh this effect is absent.…”

“…The C3 promoter region contains three estrogen responsive elements making it a highly estrogen sensitive gene in the rat uterus [9]. Because of this high sensitivity the low levels of estrogens still present in the body after ovariectomy seem to be sufficient to maintain an elevated baseline level of C3 mRNA expression, which could be downregulated by 7-O-PN, a feature previously detected for the pure antiestrogen fulvestrant in a 17-day uterotrophic assay [19].…”

“…MMLV reverse transcriptase (Life Technologies) and random hexamers were used for the first-strand cDNA synthesis. qRT-PCR analysis was performed in three independent runs as described elsewhere using 18SrRNA as an endogenous reference and SybrGreen1 to detect PCR products (Kretzschmar et al 2005). The amount of cDNA per PCR reaction corresponded to 65 ng of total RNA.…”
