NMR spectroscopic study of inclusion complexes of cetirizine dihydrochloride and β-cyclodextrin in solution

Abstract: Cetirizine dihydrochloride (CTZ), an antihistamine, forms two 1:1 inclusion complexes with β-cyclodextrin (β-CD) in aqueous solution as confirmed by detailed1H NMR, COSY and ROESY spectroscopic studies. The stoichiometry and overall binding constant of the complexes were determined by the treatment of1H NMR chemical shift change (Δδ) data. Most of the CTZ protons exhibited splitting in the presence of β-CD.

“…12 Peaks appeared in the broad multiplet between 7-8 ppm belong to peaks of 16 aromatic CH protons and 2 NH protons. 32 to verify the precision of the method since the percentage of relative standard deviation (RSD) was found to be below 2% which is within the targeted interval ( Table 5). 36 Meanwhile this method was specific for the determination and quantification of LCD.…”

“…Earlier results on the inclusion complex of β‐CD and cetirizine reported by Syed et al22 cannot be compared with the results presented here because the ROESY spectrum presented in Figure 4 of Syed et al22 suffers from an insufficient digital resolution prohibitive of extracting any details about the structure of the inclusion complex.…”

“…Very little has been reported in literature on the complex formation between cetirizine and CDs. So far, only nuclear magnetic resonance (NMR) studies on cetirizine–β‐CD complex22 have been performed, and as we will substantiate in the discussion section, these results are in part inconclusive. A couple of patents describe pharmaceutical formulations of cetirizine containing CDs as agents for the reduction of the unpleasant bitter taste 23,24.…”

“…We compared our result with cetirizine dihydrochloride (CTZ) which has some structural similarity with FLN. CTZ, an antihistamine drug used to treat allergies, formed 1:1 inclusion complexes in which the penetration of CTZ into the β-CD cavity was from wider rim side [ 56 ]. Similarly our 2D 1 H- 1 H ROESY and molecular docking approach together provide information about the penetration of FLN from the wider rim side of the β-CD cavity [ 56 ].…”

Moreover, F-containing aromatic ring of FLN positioned towards the narrower rim of the β-CD truncated cone, which is also observed in 2D 1 H- 1 H ROESY spectrum containing the cross peak between H-1, H-2 and H-5′.…”
