NMR Mapping of the Antigenic Determinant Recognized by an Anti-Gp120, Human Immunodeficiency Virus Neutralizing Antibody

Abstract: The 24‐amino‐acid peptide RP135 (NNTRKSIRIQRGPGRAFVTIGKIG) corresponds in its amino acid sequence to the principal neutralizing determinant of the human immunodeficiency virus type‐1, IIIB isolate (HIV‐1IIIB, residues 308– 331 of the envelope glycoprotein gp120). In order to map the antigenic determinant recognized by 0.5β, the complex of RP135 with an anti‐gp120 HIV neutralizing antibody, 0.5β, which cross reacts with the peptide, was studied by using two‐dimensional NMR spectros‐copy. A combination of homonu… Show more

“…Until the early 90's, predictions of the conformation of V3 loop were based on a neural network approach which yielded for the 35-residue peptide, a -strand -reverse -turn --strand --helix structure [62]. Experimental evidence in solid state through X ray crystallography [49,63,64,[71][72][73][74][75][76][77][78][79][80][81][82][83][84][85] and in solution using NMR spectroscopy [42,67,[86][87][88][89][90][91][92][93][94][95][96][97][98][99][100][101][102][103] suggest that predictable conformational features are present in V3-derived peptide epitopes from various HIV-1 strains.…”

“…It has been found that the slow dissociation rate of HIV-neutralizing antibody complexes with gp120 is correlated to the efficiency of the virus neutralization (13). Previous 1 H NMR studies of the 0.5β Fab/PND peptide complexes indicated that Lys 5 and Ile 20 constitute the boundaries of the determinant recognized by the antibody, and competitive ELISA measurements using serial deletions of residues from the PND sequence strongly supported the NMR observations (11).…”

“…Measurements of the binding constant of 0.5β to gp120 and to the PND peptide revealed similar affinities of 2 × 10 8 and 7 × 10 7 M -1 , respectively (11,12). The dissociation rate of the 0.5β/PND peptide complex is 2 × 10 -3 s -1 at 37°C (11). It has been found that the slow dissociation rate of HIV-neutralizing antibody complexes with gp120 is correlated to the efficiency of the virus neutralization (13).…”
