NMR Derivatives for Quantification of2H and13C‐Enrichment of Human Glucuronide from Metabolic Tracers

Abstract: Quantification of 2 H and 13 C enrichment distributions in human urinary glucuronide following ingestion of 2 H 2 O and 13 C gluconeogenic tracers was achieved by NMR spectroscopy of the 1,2-O-isopropylidene-a-D-glucofuranurono-6,3-lactone and 5-O-acetyl-1,2-O-isopropylidene-a-D-glucofuranurono-6,3-lactone derivatives. The derivatization process is simple and can be applied to any glucuronide species. The derivatives are highly soluble in acetonitrile and generate well-resolved and narrow 2 H and 13 C NMR sign… Show more

“…Paracetamol glucuronide in this solution was derivatized to monoacetoneglucuronic lactone (MAGL) as previously described (14). Two thousand units of ␤-glucuronidase (H. Pomatia, Sigma Chemical, St. Louis, MO) were added and the solution was incubated at 45°C for 48 hr.…”

“…The acetic acid fractions were evaporated off, and the residue was dissolved in 0.1 mol/l acetate buffer, pH 5.0. The acetaminophen glucuronide was converted to monoacetoneglucose (MAG) by a slight modification of the method of Jones et al (21). Acetaminophen glucuronide in acetate buffer was added with glucuronidase (Helix Pomatia) and incubated for 48 h at 37°C.…”
