NMR- and LC–MS/MS-based urine metabolomic investigation of the subacute effects of hexabromocyclododecane in mice

Abstract: In the present study, both untargeted and targeted metabolomics approaches were used to evaluate the subacute effects of hexabromocyclododecane (HBCD) on mice urine metabolome. Untargeted metabolomics based on (1)H NMR showed that HBCD exposure disturbed mice metabolism in both dosed groups, especially in high dosed group. The low-dose HBCD led to a decrease in alanine, malonic acid, and trimethylamine (TMA). High-dose HBCD-treated mice developed high levels of citric acid and 2-ketoglutarate, together with de… Show more

“…Finally, the current study applied a single metabolomic platform (NMR) and therefore does not cover a wide range of metabolomes due to the diversity of physicochemical properties and the broad range of metabolite concentrations. Thus, future study with multiple metabolomics platforms (e.g., GC-MS and LC-MS) or even multiple omics technologies (e.g., proteomics and transciptomics) can be integrated to provide more comprehensive and definitive meridian-specific markers 48 50 .…”

“…Targeted and untargeted metabolomic approaches have been developed to track variations in specific metabolites of interest or for a global assessment of metabolic profiles. Data gathered from these approaches can be used to connect metabolite alterations to the modulation of metabolic pathways, leading to the elucidation of potential mechanisms of action [20,21,22]. When combined with transcriptomics, these methods can provide a global picture of potential mechanisms associated with oral carcinogenesis and oral cancer chemoprevention by BRB phytochemicals, and identify novel targets for oral cancer prevention and treatment.…”

“…In addition, the fat intake was lower in the “culm” group than in the “shoot” group, however, the serum lipid level was higher in the “culm” animals than in the “shoot” group. These data show that “culm” group pandas were in short supply of energy with depressed tricarboxylic acid cycle activity, and mobilized amino acids, glycogen and lipids to compensatively trigger the tricarboxylic acid cycle to meet their energy requirements 29 31 . Thus, culm pandas were in trend of negative energy balance.…”
