Abstract: Mitophagy is an emerging paradigm for mitochondrial quality control and cell homeostasis. Dysregulation of mitophagy can lead to human pathologies such as neurodegenerative disorders and contributes to the aging process. Complex protein signaling cascades have been described that regulate mitophagy. We have identified a novel lipid signaling pathway that involves the phospholipid cardiolipin (CL). CL is synthesized and normally confined at the inner mitochondrial membrane. However, upon a mitophagic trigger, i… Show more

“…It is possible that the amplified NME1/2 signal in the nucleus of irradiated cells indicates another individual function of the NME1 and/or NME2 induced by DNA damage. Local fueling of certain nuclear proteins with a specific type of nucleotides is a valid possibility, since similar actions of the NME family members have been described [24,72]. However, the massive, simultaneous movement of both NME proteins into the nucleus favors the hypothesis that they are both engaged in the same function connected to DNA damage repair.…”

“…Ten human genes (NME1–10) encode isoforms of NME [ 2 , 3 ] where the gene products differ in their functions and subcellular localizations (for a review see [ 54 ]). Structures are available for NME1 (1jxv, 1ucn, 2hvd, 2hve, 3l7u, 4eno, and 5ui4), NME2 which is also a transcription factor (1nsk, 1nue, 3bbb, 3bbc, and 3bbf), NME3 involved in DNA repair (1zs6), the mitochondrial NME4 (1ehw) that interacts with cardiolipin-containing membranes [ 55 ], and FAP67 (an analog to NME7) (6ua2).…”

“…The NME4 hexamer enables intermembrane transfer of the mitochondrial phospholipid cardiolipin between inner and outer mitochondrial membrane, which can serve as a signal for mitophagy or apoptosis, depending of the cardiolipin oxidation state. The bioenergetics and lipid signaling functions seem to be mutually exclusive, making this protein a sort of bifunctional nanoswitch ( 115,116 ; Schlattner et al, 117 …”
