NLRP3 Inflammasome Pharmacological Inhibitors in Glycyrrhiza for NLRP3-Driven Diseases Treatment: Extinguishing the Fire of Inflammation

Abstract: Inflammation is the tissues’ defense response after the body is stimulated by microbial infection or damage signals, and it is initiated when pattern recognition receptors recognize pathogen-related molecular patterns and danger-related molecular patterns. The hyperactivation of NLRP3 inflammasome, the main driving force of immune outbreaks, is involved in a wide range of inflammatory diseases. Meanwhile, growing evidence has indicated that the development of NLRP3-targeted therapies offers great potential and… Show more

“…Flavonoids are considered to be the main biologically active components of licorice. Studies revealed that flavonoids exert different pharmacological functions, such as antiplatelet, antiinflammatory, anticancer, antioxidative, antimicrobial, immunomodulatory, and cardioprotective effects (14)(15)(16).…”

“…Except for MCC950, multiple drugs, such as 3,4-methylenedioxy-bnitrostyrene, bioactive derivatives (eg, echinatin, etc.) from medicinal plants, have been found to interfere directly with the NLRP3 (91,103). NLRP3-targeted therapies provide potential and prospects for the treatment of related diseases including severe asthma.…”

“…Emerging immunological studies also show that NLRP3 inflammasomes play an important role in the development of FLD and might be a promising therapeutic target for the treatment of FLD. Moreover, a variety of Chinese herbal formulations, TCM extracts, and natural products exert a wide range of anti-inflammatory effects by inhibiting the activation of the NLRP3 inflammasome and showing a potent and effective effect in various FLD ( Fan et al, 2020 ; Wang et al, 2022 ). In this review, we systematically summarized the role and mechanisms of NLRP3 inflammasome activation in FLD and how the TCM targets and regulates NLRP3 inflammasome to improve the development of FLD.…”
