NLRP3 inflammasome as a potential treatment in ischemic stroke concomitant with diabetes

Abstract: The NLRP3 (nucleotide-binding oligomerization domain-like receptor [NLR] family pyrin domain-containing 3) inflammasome is a member of the NLR family of innate immune cell sensors. These are crucial regulators of cytokine secretions, which promote ischemic cell death and insulin resistance. This review summarizes recent progress regarding the NLRP3 inflammasome as a potential treatment for ischemic stroke in patients with diabetes, two complicated diseases that often occur together. Stroke worsens glucose meta… Show more

“…The best-known inflammasome is NLRP3 made of NLRP3 protein, ASC adaptor protein (apoptosis-associated speck-like protein containing a carboxy-terminal CARD) and procaspase-1. [29][30][31] Activation of the NLRP3 inflammasome takes place in two stages: stage one (starting state)preparation (signal 1) and stage twoactivation stage (signal 2).…”

“…Targeting against the assembly and activity of the NLRP3 inflammasome is a potential and novel therapy for inflammasome-associated diseases, especially for cardiovascular diseases [55][56][57][58][59][60]. Some studies have indicated that blocking S194 phosphorylation can prevent NLRP3 inflammasome activation.…”

“…Inflammasomes are polymorphic complexes formed by pattern recognition receptors activated by various physiological or pathogenic stimuli, which makes them an important component of the innate immune response with the ability to clear pathogens and damaged cells [7]. The NLRP3 (nucleotide-binding and oligomerization domain-like receptor family pyrin domain-containing, NLRP) inflammasome is an innate immune cell sensor that belongs to the NLR family [8]. The NLRP3 inflammasome is considered to be the most characteristic and contains the sensor molecule NLRP3, an apoptosis-associated speck-like protein containing a caspase recruitment domain (CARD) (ASC), and pro-caspase-1 [9].…”
