Nkx2-5 defines distinct scaffold and recruitment phases during formation of the cardiac Purkinje fiber network

Abstract: The ventricular conduction system coordinates heartbeats by rapid propagation of electrical activity through the Purkinje fiber (PF) network. PF share common progenitors with contractile cardiomyocytes, yet the mechanisms of segregation and PF network morphogenesis are unknown.Using genetic fate mapping and temporal clonal analysis we identify cardiomyocytes committed to the PF lineage as early as E7.5. A polyclonal PF network emerges by progressive recruitment of conductive precursors to this scaffold from a … Show more

“…To image the CCS subpopulation within the heart with improved spatiotemporal resolution, advanced projection modalities in combination with genetically targeted models and optical clearing techniques provided a new possibility. The contribution of various cell lineages to the final CCS morphology was studied using genetic fate mapping (Choquet et al, 2020;Mohan et al, 2018). Employing the model expressing Cx40:GFP, whole ventricular conduction system up to Purkinje fibers was reconstructed in BABB cleared heart (Miquerol et al, 2010).…”
