Abstract: A mesophilic, neutrophilic and aerobic, ammonia-oxidizing archaeon, strain EN76T, was isolated from garden soil in Vienna (Austria). Cells were irregular cocci with a diameter of 0.6–0.9 µm and possessed archaella and archaeal pili as cell appendages. Electron microscopy also indicated clearly discernible areas of high and low electron density, as well as tubule-like structures. Strain EN76T had an S-layer with p3 symmetry, so far only reported for members of the Sulfolobales. Crenarchaeol was the major core l… Show more

“…Ammonium consumption and nitrite production were measured over a period of 6-8 days using standard colorimetric assays (Clesceri et al, 1998) and N 2 O was measured via GC (AGILENT 6890N, Vienna, Austria; injector: 120°C, detector: 350°C, oven: 35°C, carrier gas: N 2 ) in connection with an automatic sample-injection system (DANI HSS 86.50, Head-space-Sampler, Sprockhövel, Germany). Detailed sampling and sample preparation has been described previously (Stieglmeier et al, 2014a).…”

“…Strains and cultivation N. viennensis strain EN76 T was maintained at 37°C in 50 ml freshwater medium (FWM) supplemented with 2 mM NH 4 Cl, 0.5 mM sodium pyruvate and 50 μg/ml carbenicillin and buffered with HEPES (Tourna et al, 2011;Stieglmeier et al, 2014a) and inoculated at 4% v/v. Cultures were grown in Wheaton bottles (150 ml) sealed with caps inlaid with grey butyl rubber stoppers.…”

“…In both clone libraries (MJBA, MJDA) from the surface associated samples, representatives of "Candidatus Nitrososphaera gargensis" (MJBA: 35.48%, MJDA: 16.7%), Nitrososphaera viennensis (Stieglmeier et al, 2014) (MJBA: 32.3%, MJDA: 46.6%), and "Candidatus Nitrosoarchaeum limnia" (MJBA: 29.03%, MJDA: 23.3%) were the most abundant.…”

“…Thaumarchaeotal groups 1.1b and 1.1c were recently estimated to account for 76 ± 33% of total archaea in global soil samples (Auguet et al, 2010), suggesting they may have key roles in Earth's biogeochemical cycles. Currently, all cultured members of the Thaumarchaeota are autotrophic ammonia oxidizing archaea, belonging to group 1.1a and 1.1b (Konneke et al, 2005;de la Torre et al, 2008;Hatzenpichler et al, 2008;Tourna et al, 2011;Stieglmeier et al, 2014). The carbon metabolism and energy sources used by group 1.1c remain unknown.…”
