Nitrogenase in the archaebacterium Methanosarcina barkeri 227

Abstract: The discovery of nitrogen fixation in the archaebacterium Methanosarcina barkeri 227 raises questions concerning the similarity of archaebacterial nitrogenases to Mo and alternative nitrogenases in eubacteria. A scheme for achieving a 20-to 40-fold partial purification of nitrogenase components from strain 227 was developed by using protamine sulfate precipitation, followed by using a fast protein liquid chromatography apparatus operated inside an anaerobic glove box. As in eubacteria, the nitrogenase activity… Show more

“…In accordance with the results of Lob0 [38], who demonstrated that yeast mutants with only minute amounts of aldolase enzymatic activity grow well on ethanol as the sole carbon source, this confirms the presumption that several glycolytic enzymes are expressed a t a much higher level than actually required under the respective nutritional conditions. The high abundance of these enzymes ensures that these reactions d o not become rate-limiting steps in glycolysis.…”

“…Yeast mutant cells deficient in aldolase and their isogenic wild-type strain MC3 (28) were grown overnight at 30°C in a shaking incubator. The cultures were diluted 1:10 and grown for 3 h. B5 fixative was added to a final concentration of 3% formaldehyde, and incubation was continued for 10 min.…”
