Nitrogen partitioning in oak leaves depends on species, provenance, climate conditions and soil type

Abstract: Climate-tolerant tree species and/or provenances have to be selected to ensure the high productivity of managed forests in Central Europe under the prognosticated climate changes. For this purpose, we studied the responses of saplings from three oak species (i.e. Quercus robur, Q. petraea and Q. pubescens) and provenances of different climatic origin (i.e. low or high rainfall, low or high temperature habitats) with regard to leaf nitrogen (N) composition as a measure of N nutrition. Saplings were grown in mod… Show more

“…The extraction of amino acids from plant material was conducted after the method of Winter et al [58] and Hu et al [59] with slight modifications. Approximately 50 mg of frozen leaf or 80 mg root powder were mixed with 1 mL methanol/chloroform (Merck KGaA, Darmstadt, Germany) (3.5/1.5, v/v) and 0.2 mL Hepes buffer (containing 20 mM Hepes (Merck KGaA, Darmstadt, Germany), 5 mM EGTA (Sigma-Aldrich Inc., St. Louis, MO, USA), 10 mM NaF (Sigma-Aldrich Inc., St. Louis, MO, USA)).…”

“…In many forest species, organic solutes, specifically soluble carbohydrates and free amino acids, are the principal compounds involved in osmotic adjustment (Patakas et al, 2002). At the molecular level, most of these responses depend on proteins, therefore comparison of the protein profile of tolerant and sensitive populations can provide valuable information to predict the performance of a given genotype under drought conditions (Hu et al, 2012). …”

“…For this process, the mixture underwent ultrasonic extraction for 1 h; the extract was centrifuged at 13,200 r/min for 10 min at 4 °C. The supernatant was measured by the HPLC-MS method (Hu et al 2013). Amino acid content was determined by BEH Amide, 1.7 mu M, 100 × 2.1 mm column (liquid phase model UltiMate 3000, Thermo Scientific™ production; mass spectrometry model is Q Exactive™ production).…”
