Nis1 encoded by YNL078W: a new neck protein of Saccharomyces cerevisiae.

Abstract: An uncharacterized gene, YNL078W, was isolated by the two-hybrid screening method using SHS1 (one of the septin genes) as bait and designated NIS1 (Neck protein Interacting with Septins). Nis1 interacts with all septins in the two-hybrid assay system. Physical interaction between Nis1 and Shs1 in vivo was confirmed by a co-immunoprecipitation experiment. Neither disruption nor overexpression of NIS1 caused a prominent phenotypic change. NAP1 was isolated by two-hybrid screening using NIS1 as bait. We detected … Show more

“…Nis1p localizes to the bud neck and interacts with the septins, which are the most abundant sumoylated proteins in the yeast cell (Iwase and Toh-e 2001). Ris1p, which has been previously implicated in silencing, contains a RING motif, suggesting that it may have E3 ligase activity (Zhang and Buchman 1997;Hannich et al 2005).…”

“…In our previous work, we demonstrated that Nis1 interacts with septins by two-hybrid assay using Shs1 as bait (Iwase and Toh-e, 2001). During our attempt on functional analysis of Nis1, we performed a two-hybrid screening to find the Nis1 binding proteins.…”

“…In our previous work, Nis1 was isolated by a two-hybrid screening using Shs1 as bait, and was found to be localized to the bud neck in M phase and was able to bind with all septins, Nap1, Gin4 and Kcc4 (Iwase and Toh-e, 2001). Thus, Nis1 was suggested to have some relation with the mitotic signaling network, but the exact nature of this function of Nis1 has yet to be fully elucidated.…”

“…Microscopic observations were made according to the method described by Yoshida and Toh-e (2001) and Iwase and Toh-e (2001). Cells in logarithmic phase were harvested and fixed with 3.7% formalin at a room temperature for 30 minutes.…”
