Nicotine protects against DSS colitis through regulating microRNA-124 and STAT3

Abstract: Nicotine upregulates miR-124 expression in ulcerative colon tissues and cells. MiR-124 is required for the protective role of nicotine in DSS colitis mice and epithelial cells. The protective effect of nicotine in murine DSS colitis depends on blocking STAT3 activation. MiR-124 mediates the inhibitory role of nicotine on STAT3/p-STAT3. Targeting miR-124 and STAT3 represents a novel approach for treating ulcerative colitis.

“…Furthermore, subcutaneous or oral nicotine administration to rats treated with DNBS led to, respectively, decreased or increased colon inflammation [58,59]. Finally, while oral or subcutaneous nicotine administration attenuated inflammation caused by DSS treatment in mice [50,80], intraperitoneal nicotine injection had no effects [81,82]. Inconsistencies related to different routes of administration of CS components were also observed in human studies [83][84][85][86].…”

“…However, differences in immune cell recruitment [48], cytokine secretion [49][50][51], mucosal barrier [52,53] and oxidative stress were observed [54,55], although without evident tissue damage. Consistent with results of human epidemiological studies, CS had opposing effects on development of CD (negatively) and UC (positively) in several, but not all, of their respective IBD models.…”

“…The DAI scoring system described previously (17,19) consists of body weight loss (zero, none; one, 1-5%; two, 5-10%; three, 10-15%; four, 15-20%), stool consistency (zero, normal; two, loose stools; four, watery diarrhea), and bloody stools (zero, normal; two, slight bleeding; four, gross bleeding). The sum of the three values constitutes the DAI.…”
