Nicotinamide N-methyltransferase is overexpressed in prostate cancer and correlates with prolonged progression-free and overall survival times

Abstract: Nicotinamide N-methyltransferase (NNMT) has been identified to be associated with tumorigenesis and the malignant transformation of numerous types of cancer. The aim of the present study was to explore the expression and prognostic significance of NNMT in prostate cancer (PCa). Immunohistochemical NNMT expression was examined in 26 cases of benign prostate hyperplasia (BPH), 18 cases of high-grade prostatic intraepithelial neoplasia (HGPIN) and 120 cases of PCa. While rarely expressed in BPH (8/26 cases; 30.8%… Show more

“…In prostate carcinoma, enzyme levels were negatively correlated with Gleason score. Contrary to what above reported for other neoplasms, NNMT expression was significantly associated with prolonged progression‐free survival and overall survival in patients diagnosed with advanced prostate cancer . Results obtained from a recent study, carried out in a cohort of patients with gastric cancer, revealed that NNMT tumour expression is positively related to primary tumour size, lymph node or distant metastasis and pathological stage.…”

“…Contrary to what above reported for other neoplasms, NNMT expression was significantly associated with prolonged progression-free survival and overall survival in patients diagnosed with advanced prostate cancer. 37 Results obtained from a recent study, carried out in a cohort of patients with gastric cancer, revealed that NNMT tumour expression is positively related to primary tumour size, lymph node or distant metastasis and pathological stage. Kaplan-Meier curves demonstrated that NNMT represents an important negative prognostic predictor of overall survival of gastric cancer patients.…”

“…NNMT is a cytosolic enzyme that catalyzes the N-methylation of NCA, pyridines and analogues using S-adenosyl-L-methionine (AdoMet) as the methyl donor. Previous studies have examined the effects of NNMT knockdown or overexpression on various cancer cell phenotypes in vitro , yielding conflicting results49505152. Some of them identified positive correlation between overexpression of NNMT and tumor cell aggressiveness5051.…”

“…NNMT expression was not correlated with increasing fat mass in HFD mice, but is elevated when mice consume HFD. Research on NNMT has largely focused on its association with carcinogenesis [28,29]. Likewise SIRT7 has also been implicated in carcinogenesis [30,31].…”
