Nicotinamide ameliorates energy deficiency and improves retinal function in Cav‐1‐/‐ mice

Abstract: Caveolin-1(Cav-1) is involved in lipid metabolism and energy homeostasis, which is important for the energetically demanding retina. Although retinal function deficits were noted in Cav-1 knockout (Cav-1 -/-) mice, the underlying causes remain largely unknown. Here, we investigate if the disruption in energy homeostasis presents a potential mechanism for retinal function deficits in Cav-1 -/retina and if it can be ameliorated by nicotinamide (NAM). In this study, NAM was administrated orally for 2 weeks in Cav… Show more

“…Free fatty acids are also one of the substances that contribute to oxidative stress ( Jiang and Yan, 2022 ). Oxidized lipids are metabolites of polyunsaturated fatty acids (arachidonic acid, linoleic acid, alpha-linolenic acid) that undergo auto-oxidation or are generated by the action of specific enzymes ( Tang et al, 2021 ). 20-COOH-AA is produced by a series of enzymatic oxidation reactions of arachidonic acid.…”

“…The ERG is a common and sensitive measurement used to evaluate retinal function ( Tang et al, 2021 ). The a-wave provides information associated with the photoreceptors (cones and rods) and b-waves regarding the physiology of bipolar and Müller cells ( Scalabrino et al, 2015 ).…”

“…This feedback may lead to energy deficiency and further deteriorate neurodegeneration, which is also reported to be the key pathogenesis of glaucoma. Thus, targeting energy deficiency might be another therapeutic perspective for glaucoma ( Williams et al, 2017 ; Tang et al, 2021a ).…”
