NGS in the clinical microbiology settings

Abstract: We hypothesized that targeted NGS sequencing might have an advantage over Sanger sequencing, especially in polymicrobial infections. The study included 55 specimens from 51 patients. We compared targeted NGS to Sanger sequencing in clinical samples submitted for Sanger sequencing. The overall concordance rate was 58% (32/55) for NGS vs. Sanger. NGS identified 9 polymicrobial and 2 monomicrobial infections among 19 Sanger-negative samples and 8 polymicrobial infections in 11 samples where a 16S gene was identif… Show more

“…Beside LA, a variety of technologies are revolutionizing clinical microbiology. These include MALDI-TOF MS ( Seng et al., 2009 ), time-lapse microscopy for ID and phenotypic AST ( Charnot-Katsikas et al., 2018 ), molecular diagnostic tests and syndromic panels ( Arena et al., 2017 ; Trotter et al., 2019 ), and next-generation sequencing ( Mitchell and Simner, 2019 ; Pitashny et al., 2022 ). All of them can significantly improve the diagnosis and therapy of infections, but as stated above, they are primarily complementary to culture ( Trotter et al., 2019 ).…”

“…Also, in the realm of thoracic oncology, Polymerase Chain Reaction (PCR) and Next-Generation Sequencing (NGS) studies serve as prominent tools for identifying cancer cells and deciphering the genetic alterations driving cancer growth [17,18]. PCR-based techniques exhibit exceptional efficacy in detecting minute quantities of circulating tumor DNA (ctDNA), facilitating the early identification of tumor presence.…”
