NGF Controls Axonal Receptivity to Myelination by Schwann Cells or Oligodendrocytes

Abstract: Axons dictate whether or not they will become myelinated in both the central and peripheral nervous systems by providing signals that direct the development of myelinating glia. Here we identify the neurotrophin nerve growth factor (NGF) as a potent regulator of the axonal signals that control myelination of TrkA-expressing dorsal root ganglion neurons (DRGs). Unexpectedly, these NGF-regulated axonal signals have opposite effects on peripheral and central myelination, promoting myelination by Schwann cells but… Show more

“…OPCs were purified from 6 to 7 day old (P6-P7) rat brain cortices with a panning protocol adapted from one previously described (2). Briefly, Petri dishes containing a goat anti-mouse IgGϩIgM secondary antibody solution (Jackson Laboratories) were incubated overnight.…”

“…The in vitro myelinating coculture represents a reduced system that is ideal for manipulating the microenvironment (15). Similar to oligodendroglial development in vivo, oligodendrocyte precursor cells (OPCs) cocultured with purified sensory neurons undergo an initial period of proliferation in response to axonal cues (15,16) before differentiating into myelinating oligodendrocytes.…”

“…In vitro study have indicated that NGF promotes myelination in Schwann cells and inhibits conversion of premyelinating into myelinating oligodendrocyte in a cellular system derived from embryonic dorsal root ganglia (53). However, it is unknown whether the same observations are applicable to remyelination after injury.…”
