NFE2 Induces miR-423-5p to Promote Gluconeogenesis and Hyperglycemia by Repressing the Hepatic FAM3A-ATP-Akt Pathway

Abstract: Hepatic FAM3A expression is repressed under obese conditions, but the underlying mechanism remains unknown. This study determined the role and mechanism of miR-423-5p in hepatic glucose and lipid metabolism by repressing FAM3A expression. miR-423-5p expression was increased in the livers of obese diabetic mice and in patients with nonalcoholic fatty liver disease (NAFLD) with decreased FAM3A expression. miR-423-5p directly targeted FAM3A mRNA to repress its expression and the FAM3A-ATP-Akt pathway in cultured … Show more

“…FAM3A is a new mitochondrial protein that enhances ATP production, while other members of FAM3 gene family are secretory proteins . FAM3A suppresses hepatic gluconeogenesis and lipogenesis by activating protein kinase B (Akt) and adenosine 5′‐monophosphate (AMP)‐activated protein kinase (AMPK) pathways, and a decrease in hepatic FAM3A expression causes fasting hyperglycemia and hepatic lipid deposition . FAM3A is a target gene of peroxisome proliferator‐activated receptor gamma (PPARγ), and it mediates the beneficial effects of PPARγ’s activation on liver ischemia/reperfusion injury .…”

“…It inhibits tumour growth by inducing cell cycle arrest and apoptosis in the early stage of tumour formation. 7,8 It has been reported that Akt activation plays a critical role in TGFβ-promoted development of breast cancer. 3,4 Protein kinase B (Akt) is an important intracellular molecule that stimulates cell proliferation and inhibits apoptosis 5,6 beyond its roles in regulating glucose and lipid metabolism as the key molecule of insulin signalling transduction.…”

“…Eight‐week‐old male C57BL/6 mice and 8‐ to 12‐week‐old male db/db and db/m (C57BKS background Jackson Laboratory, USA) were used in this study. 8‐10 week old male C57BL/6 mice were fed on 45% high fat diet (HFD) or normal diet (ND) for 12 weeks to induce diabetic and steatotic phenotype as detailed previously . 12‐ to 16‐ week old male db/db mice and db/m mice on BKS ground were also used in this study.…”
