NF2 deficiency accelerates neointima hyperplasia following vascular injury via promoting YAP-TEAD1 interaction in vascular smooth muscle cells

Abstract: Neurofibromin 2 (NF2), a potent tumor suppressor, is reported to inhibit proliferation in several cell types. The role of NF2 in neointima hyperplasia after vascular injury is unknown. We explored the role of NF2 in proliferation, migration of vascular smooth muscle cell (VSMC) and neointima hyperplasia after vascular injury. NF2 phosphorylation was elevated in VSMC subjected to platelet-derived growth factor (PDGF)-BB and in artery subjected to vascular injury. Mice deficient for Nf2 in VSMC showed enhanced n… Show more

“…To explore whether PIKfyve affects the proliferation and migration of VSMCs in vivo, we assessed the effect of PIKfyve in an established model of neointima hyperplasia induced by vascular wire injury. 34 To analyze the role of PIKfyve in vascular intima hyperplasia, qPCR and Western blot were used to detect the content of PIKfyve in carotid artery. After 28 days of injury, we observed increased mRNA and protein level of PIKfyve in vessels (Figs.…”

“…To explore the potential role of VRK1 in the proliferation and migration of VSMCs, we firstly examined VRK1 expression in synthetic VSMCs. Platelet-derived growth factor (PDGF)-BB is known to promote phenotypic transition of VSMC ( 10 ). In the current study, VRK1 was upregulated by PDGF-BB stimulation in both dose-dependent manner ( Fig.…”

“…C57BL/6J mice (Dashuo Animal Science and Technology, Chengdu, Sichuan) were raised as following described: room temperature, 12-hour light/dark cycle, periodic air changes and free access to food and water. Establishing vascular injury model was performed as described before ( 10 ). Briefly, mice were firstly anesthetized and received a small midline incision in the neck area.…”

“…2020ky014; Chengdu, Sichuan, China). Injury model of common carotid artery was established as described in our previous literature [ 18 ]. Briefly, after anaesthetization with pentobarbital (40 mg/kg, intraperitoneal), a small midline incision in the neck area was conducted.…”
