NF-κB Family of Transcription Factors: Central Regulators of Innate and Adaptive Immune Functions

Abstract: SUMMARY Transcription factors of the Rel/NF-κB family are activated in response to signals that lead to cell growth, differentiation, and apoptosis, and these proteins are critical elements involved in the regulation of immune responses. The conservation of this family of transcription factors in many phyla and their association with antimicrobial responses indicate their central role in the regulation of innate immunity. This is illustrated by the association of homologues of NF-κB, and thei… Show more

“…However, NF-B family members can also influence multiple DC functions, including expression of costimulatory molecules and proinflammatory cytokines (19). To test whether the requirement for NF-B1 expression in Th2 cell differentiation was intrinsic to the T cell lineage or whether NF-B1 can regulate APC function during a Th2 response, an adoptive transfer model in which purified NF-Bsufficient OVA-specific DO11.10 TCR Tg CD3 ϩ T cells were transferred into syngeneic WT or NF-B1 Ϫ/Ϫ recipients was used.…”

“…This form of NF-κB is in fact the most abundant and common activator of both gene activation and viral transcription, out of all the homo-and heterodimers of the Rel family [34][35][36]. In most cases, the rapid degradation of the inhibitor protein IκB-α precedes NF-κB translocation.…”

“…The downregulation by NMP on the NF-κB pathway has been rec- ognized in macrophages (Ghayor et al, 2015) and in osteoclasts . And the NF-κB pathway is indicated to regulate the expression of multiple genes involved in inflammatory and immune response (McKay and Cidlowski, 1999;Caamaño and Hunter, 2002). And the IKK (inhibitory κB (IκB) kinase)-mediated phosphorylation-induced proteasomal degradation of IκB inhibitor, enabling the active NF-κB to translocate to the nucleus (Vallabhapurapu and Karin, 2009).…”
