Next-generation derivatization reagents optimized for enhanced product ion formation in photodissociation-mass spectrometry of fatty acids

Abstract: Ultraviolet-photodissociation (UVPD) mass spectrometry is an emerging analytical tool for structural elucidation of biomolecules including lipids. Gas phase UVPD of ionised fatty acids (FAs) can promote fragmentation that is diagnostic...

“…This signature is used in the MS 3 PD-CID experiments for locating C=C in unsaturated lipids. It is worth noting that RDD from the allylic position of the C=C is a prominent fragmentation channel directly resulting from 266 nm UVPD of the aryl-iodine derivatized FAs ( 49 ), which facilitates the analysis of C=C location isomers in a mixture of 37 FAs implemented on an LC-MS platform ( 50 ).…”

“…Combining the analytical benefits of spectroscopy and mass spectrometry, the resulting action spectra reveal absorption events that produce a specified molecular mass change, 41−45 which can guide the design of new substrates with desirable photodissociation profiles. 46 We show that the efficiency of nitrile imine formation as a function of irradiation wavelength can be readily surveyed in the gas phase, independent of dipolarophile cycloaddition. Moreover, we demonstrate that this mass spectrometry-based approach is compatible with crude mixtures of macromolecules, enabling characterization and evaluation of tetrazole performance prior to laborious purification procedures.…”

“…However, the location of C=C in FAs is typically not obtained from these approaches. Alternative ion activation methods which can provide detailed structural information have been developed, including charge-remote fragmentation (17,18), ozone-induced dissociation (OzID) (19), ultraviolet photodissociation (UVPD) (20,21), ion/ion reactions (22), etc. On the other hand, the combination of functional group specific derivatization and MS 2 CID greatly expands the capability of detailed structural analysis of lipids on commercial MS platforms.…”
