New Thermal Neutron Capture Therapy for Malignant Melanoma: Melanogenesis‐Seeking 10B Molecule‐Melanoma Cell Interaction From In Vitro to First Clinical Trial

Abstract: Human melanoma regression by single thermal neutron capture therapy (NCT) using melanoma-seeking 10B compounds has been achieved. Since 1972, the aim of my team has been to synthesize tumor-seeking 10B-compounds possessing selective affinity for specific metabolic activity of the target cancer cells. Once the melanoma takes up these 10B compounds, thermal neutrons, which cause insignificant cell damage, are easily absorbed by nonradioactive 10B, inducing the 10B(n, alpha)7Li reaction and releasing the high LET… Show more

“…Other than patients with primary brain tumors, the second largest group that has been treated by BNCT were those with cutaneous melanomas. Mishima et al previously had carried out extensive studies in experimental animals with either primary or transplantable melanomas using 10 B-enriched BPA as the capture agent (175,176). The use of BPA was based on the premise that it would be selectively taken up by and accumulate in neoplastic cells that were actively synthesizing melanin (177).…”

“…Better "B-containing substances have now been developed and promising approaches have recently been reported for the treatment of both malignant gliomas ( 5-9) and melanomas (10). The strategies for developing new boron-containing substances have been reviewed ( 1 I , 12) and were recently discussed at an international workshop held in Uppsala in May 1991.…”

“…2b). Promising BNCT studies on experimental animals have been reported ( 10,53) and the first clinical trial with BPA was performed by Mishima (10,36,54). The first patient suffered from an inoperable malignant melanoma lesion on the left occiput and the perilesional injections of BPA, followed by irradiation with thermal neutrons, was successfully tested.…”

“…9)11) It exhibits two crystal structures, orthorhombic and hexagonal, which differ significantly in coordination number, lattice volume, etc. While orthorhombic GdF 3 (space group Pnma) is thermodynamically stable at room temperature as shown in Table 1, there are very few studies on unstable hexagonal GdF 3 (space group p3-c1).…”
