Surface chemistry of gold nanoparticles determines the biocorona composition impacting cellular uptake, toxicity and gene expression profiles in human endothelial cells

This study investigated the role of nanoparticle size and surface chemistry on biocorona composition and its effect on uptake, toxicity and cellular responses in human umbilical vein endothelial cells (HUVEC), employing 40 and 80 nm gold nanoparticles (AuNP) with branched polyethyleneimine (BPEI), lipoic acid (LA) and polyethylene glycol (PEG) coatings. Proteomic analysis identified 59 hard corona proteins among the various AuNP, revealing largely surface chemistry-dependent signature adsorbomes exhibiting human serum albumin (HSA) abundance. Size distribution analysis revealed the relative instability and aggregation inducing potential of bare and corona-bound BPEI-AuNP, over LA-and PEG-AuNP. Circular dichroism analysis showed surface chemistry-dependent conformational changes of proteins binding to AuNP. Timedependent uptake of bare, plasma corona (PC) and HSA corona-bound AuNP (HSA-AuNP) showed significant reduction in uptake with PC formation. Cell viability studies demonstrated dose-dependent toxicity of BPEI-AuNP. Transcriptional profiling studies revealed 126 genes, from 13 biological pathways, to be differentially regulated by 40 nm bare and PC-bound BPEI-AuNP (PC-BPEI-AuNP). Furthermore, PC formation relieved the toxicity of cationic BPEI-AuNP by modulating expression of genes involved in DNA damage and repair, heat shock response, mitochondrial energy metabolism, oxidative stress and antioxidant response, and ER stress and unfolded protein response cascades, which were aberrantly expressed in bare BPEI-AuNP-treated cells. NP surface chemistry is shown to play the dominant role over size in determining the biocorona composition, which in turn modulates cell uptake, and biological responses, consequently defining the potential safety and efficacy of nanoformulations. Abstract:The Magnetic Resonance Imaging (MRI) is a noninvasive technique based on the magnetic properties of the 1 H atoms. Currently, about 30% of MRI exams use paramagnetic compounds, which are defined as contrast agents (CAs), able to reduce the relaxation times (T 1 and T 2 ) of hydrogen atoms from water molecules. In this review, we show the main compounds employed as CAs. Among these, Gd 3+ complexes are frequently used, but problems related to their high toxicity and relaxivities, promoting the development of new ACs, such as iron oxides, hybrid compounds as well as Mn 2+ and Ni 2+ complexes, which are less toxic and more effective.Keywords: Contrast agents; MRI; paramagnetic compounds. ResumoA Imagem por Ressonância Magnética (IRM) é uma técnica não invasiva baseada nas propriedades magnéticas dos átomos de 1H. Atualmente, cerca de 30% dos exames de IRM utilizam compostos paramagnéticos capazes de reduzir os tempos de relaxação (T 1 e T 2 ) dos átomos de hidrogênio a partir das moléculas de água, que são definidos como agentes de contrastes (ACs). Nesta revisão, mostramos os principais compostos utilizados como ACs. Entre eles, complexo de Gd3+ são frequentemente utilizados, mas os problemas relacionados com a sua elevada toxicidade e relaxatividade, promoveu o desenvolvimento de novos ACs, tais como óxidos de ferro, compostos híbridos, bem como complexos de Mn2+ e Ni2+ que são menos tóxicos e mais eficazes.

Nanoscience is the branch of science pertaining to the study of minute particles on an atomic or molecular scale, whose size is measured in nanometers. A nanometer represents one billionth of a meter (i.e., 10 -9 m). Nanotechnology is an emerging branch of engineering that involves the use of particles on a nanoscale (1-100 nm). Thus, the application of nanotechnology in the field of dermatology is the Nanodermatology. Nanodermatology represents one of the most emerging fields for which an increasing interest is rising among scientists as well as pharmaceutical companies. Nanotechnology has revolutionized the treatment of several skin diseases. It is effective in targeted delivery of active medicaments and cosmetic ingredients. The skin forms the first point of contact for a diverse number of nanomaterials. Nanomedicine in dermatology has opened a new era in the diagnosis and treatment of various skin disorders. Possible applications of nanotechnology in dermatology and cosmetics include sunscreens, moisturizers, anti-aging formulations, phototherapy, anti-sepsis, vaccines, skin cancers, hair and nail care, etc. In this review after an introduction of nanotechnology, authors have described various types of nanoparticles followed by various possible indications of nanotechnology in dermatology and cosmetics. An account on safety of nanoparticles has also been added to the review. properties. Various other agents including vitamins (Vit. A, E, and K) and certain antioxidants like Carotenoids, lycopene, etc have been introduced in liposomes. These increase the physical and chemical stability of liposomes when dispersed in water [12]. Due to their biocompatible, biodegradable and nontoxic nature, liposomes find uses in a variety of cosmeceuticals as they encapsulate active moiety easily [11]. When associated to medication; liposomes ensure a high penetration into hair follicles [13]. Solid lipid nanoparticles (SLN)SLNs are colloidal carriers with size varying from 50 nm to 1000 nm. In these particles, Lipids (physiological lipids) are dispersed in an aqueous solution of surfactant or water [14]. SLNs possess various favourable properties viz., a) Presence of physiological and biodegradable lipids with low toxicity b) Occlusive properties which increase skin moisture level, c) Having a small size which ensures increased penetration of active ingredients, and d) Possessing characteristics of physical sunscreens of their own. These properties have made them popular in cosmetics [15,16]. SLNs have also been employed as topical vehicles for perfumes. When perfumes or fragrances are incorporated in SLNs, their release is delayed giving a prolonged effect of the perfume [17].
