New spectrofluorimetric analysis of dapagliflozin after derivatization with NBD‐Cl in human plasma using factorial design experiments

Abstract: A new validated spectrofluorimetric method was proposed for dapagliflozin (DGF) analysis in bulk, plexin its commercially available tablets and in spiked human plasma. The proposed spectrofluorimetric method depended on the formation of a fluorescent complex soluble in organic liquids by a substitution reaction between 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) reagent and DGF in aqueous buffered solution at pH 7. The fluorescence intensity was measured at 522 nm after excitation at 453 nm. The high se… Show more

“…The application of the AQbD concept throughout the development and validation of analytical methods has a considerable positive effect on the method life cycle. [49,50] For the final method optimization, the four critical method attributes (CMAs), i.e. pH of the TSB, heating temperature, and concentrations of ninhydrin and phenylacetaldehyde, were selected.…”

“…It is a great tool for a variety of chemical and biological applications due to its amazing fluorescence capabilities. NBD-Cl exhibits a typical nucleophilic substitution mechanism when interacting with various functionality groups [14][15][16], producing stable NBD-labeled products. These derivatives exhibit strong green fluorescence upon excitation, which facilitates their easy detection and quantification in complicated samples.…”

“…Also, the reported electrochemical methods always need electrode pre-modification. Thus, we found it urgent to establish a simple and time-saving approach for NTZ determination [19][20][21][22][23][24]. Fluorimetry is a sensitive and relatively selective approach that has been applied for NTZ analysis.…”
