New sesquiterpenoids with COX-2 inhibitory activity from the medical plant Physalis. alkekengi L. var. franchetii

Abstract: Phytochemical investigations on Physalis. alkekengi L. var. franchetii, a widespread traditional Chinese medicine, led to the isolation and identification of three new sesquiterpenoids physalisitins A-C (1-3). Their structures were elucidated by NMR and HRESIMS analysis, and their absolute configurations were determined by quantum chemical NMR and ECD calculations, as well as by comparing their optical rotation values with those known analogues. All of the isolated compounds were evaluated for their cyclooxyge… Show more

“…This has the effect of promoting the ubiquitous degradation of mesenchymal-epithelial transition protein (MET) to inhibit the PI3K/Akt/mTOR pathway, which ultimately inhibits the proliferation, migration, and invasion of gastric cancer cells. In addition to this, Zhang et al [ 85 ] also found that sesquiterpenoids have some cytotoxic properties. In conclusion, P. alkekengi has certain inhibitory effects on a variety of tumours, and its mechanism may be related to inducing G2/M phase arrest of cancer cells through various pathways to promote apoptosis and inhibit the proliferation of cancer cells.…”

“…Chen et al [ 84 ] found that the combination of physalin A, luteolin, and cynaroside had significant synergistic inhibitory effects on LPS-induced NO and TNF-α release from macrophages, and the combination significantly reduced LPS-induced expression of iNOS protein. In addition, Zhang et al [ 85 ] found a dose-dependent inhibition of COX-2 enzymes by sesquiterpenoids in P. alkekengi , and Xu [ 15 ] showed that withanolides also have strong anti-inflammatory activity. In conclusion, numerous studies have shown that all extracted parts of P. alkekengi show significant anti-inflammatory activity and can be widely studied and applied as anti-inflammatory agents.…”

“…For example, the IC 50 values for physalisitins A, B, and C, three new sesquiterpenoids isolated from the medical plant Physalis alkekengi L. var. franchetii, were (3.22 ± 0.25), (6.35 ± 0.84), and (11.13 ± 1.47) μmol/L, respectively [36] . Furthermore, Nelumbo nucifera leaf extract was reported to show COX-2 inhibitory activity with an IC 50 of (8.69 ± 0.76) μg/mL [18] .…”

“…In recent decades, PAF has been used to treat AD with satisfactory therapeutic effect. [9] Physalins are Physalisderived steroid derivatives and they exhibit strong properties both in reducing inflammation and modulating the immune system. [10][11][12][13] Tumor necrosis factor-α (TNF-α), interleukin (IL)-6, IL-12 production are all inhibited by Physalin F, as is nuclear factor kappa-B (NF-kB), a crucial inflammatory transcription factor.…”

“…Traditional Chinese medicine's comprehensive approach has advantages in enhancing cognitive function in AD via numerous biochemical pathways and may have the capacity to prevent AD. In recent decades, PAF has been used to treat AD with satisfactory therapeutic effect [9] . Physalins are Physalis‐derived steroid derivatives and they exhibit strong properties both in reducing inflammation and modulating the immune system [10–13] .…”
