New separation method for monoclonal immunoradiometric assays and its application to assays for thyrotropin and human choriogonadotropin.

Abstract: We describe a novel separation procedure for immunoradiometric assays involving monoclonal antibodies in which both radiolabeled and capture antibodies are used in solution, the capture antibody being labeled with fluorescein isothiocyanate (FITC). Separation is achieved by incubation with anti-FITC antibodies on magnetic particles. This technique enhances reaction kinetics relative to those of assays in which a solid-phase capture antibody is used, thus allowing faster reaction times and more economic use of … Show more

“…Analysis of serum PRL and CORT concentrations was carried out blind to patient status in the Department of Clinical Biochemistry, King's College Hospital. The prolactin was assayed using the immunoradiometric method MAIAClone from Serano (Hampshire, UK) described by Rattle et al (1984), and CORT was assayed using the Coat-A-Coat radioimmunoassay from DPC (California, USA). The inter-assay coefficient of variance was less than 6% for both assays.…”

“…The results are expressed as mi.u./ml of standards calibrated against the WHO 1st IRP for human LH (68/40). The use of high-affinity monoclonal antibodies to FSH and LH offers an increased level of sensitivity and specificity compared with traditional methods (Soos & Siddle, 1983;Rattle et al, 1984). Oestradiol-17ß was measured in serum by a direct radioimmunoassay using a kit purchased from Biodata (Serono Diagnostics Ltd).…”

“…Serum and amniotic fluid TSH concentrations were determined by a double monoclonal RIA (Rattle et al, 1984); fT4 and T3 were also determined by RIA, as previously described (Wilkins et al, 1985). Tanner's and Nellhause charts were used for assessing height and head circumference growth parameters respectively; whereas psychomotor achievement testing was performed according to Gesell and Amatruda's developmental schedules (Gesell, 1979).…”
