New Polyhydroxylated Furostanol Saponins with Inhibitory Action against NO Production from Tupistra chinensis Rhizomes

Abstract: Two furostanol saponins were obtained from the rhizomes of Tupistra chinensis Bak. Their structures were determined as 5β-furost-Δ 25(27) -en-1β,2β,3β,4β,5β,7α,22ξ,26-octaol-6-one-26-O-β-D-glucopyranoside (1) and 5β-furost-Δ 25(27) -en-1β,2β,3β,4β,5β,6β, 7α,22ξ,26-nonaol-26-O-β-D-glucopyranoside (2), on the basis of chemical and spectroscopic evidence. Both compounds displayed marked inhibitory action against NO production in rat abdomen macrophages induced by lipopolysaccharide (LPS) at 40 μg/mL.

“…Although several studies have reported furostanol saponin derivatives isolated from various plants (De Combarieu, Fuzzati, Lovati, & Mercalli, 2003;Kang et al, 2007), most of the studies concerning bioactivity have focused on antimicrobial or antiinflammatory activities of furostanol saponins (Iorizzi et al, 2002;Lee et al, 2014;Xu et al, 2007). There had been no studies on the anti-adipogenic activity of capsicosides until now.…”

“…Phytochemical investigation on T. chinensis in the past 15 years has resulted in the isolation of some steroidal sapogenins and steroidal saponins (Cai et al 2007;Guo et al 2009;Liu, Guo, Xue, Cheng et al 2012;Liu, Guo, Xue, Zhang et al 2012;Pan et al 2000aPan et al , 2000bPan et al , 2003Pan et al , 2006Pan et al , 2012Wu et al 2005;Xu et al 2007;Zou et al 2005Zou et al , 2006Zou et al , 2009). Previously, we reported the isolation and structural elucidation of several spirostanol sapogenins, flavonoids, lignans, a pregnane glycoside, a pregnane genin and other chemical constituents from the CHCl 3 fraction of methanol extract from T. chinensis rhizomes (Pan et al 2000a(Pan et al , 2000b(Pan et al , 2003(Pan et al , 2006.…”

“…Kai-Kou-Jian is a reputed folk medicine because of its power to markedly reduce carbuncles and to ameliorate pharyngitis (Zhan 1994). Some furostanol saponins isolated from T. chinensis rhizomes showed inhibition action against NO production (Xu et al 2007) and against COX-2 production ).…”
