Aim: Honokiol (HNK) is a natural compound isolated from the magnolia plant with numerous pharmacological activities, including inhibiting epithelial-mesenchymal transition (EMT), which has been proposed as an attractive target for anti-tumor drugs to prevent tumor migration. In this study we investigated the effects of HNK on EMT in human NSCLC cells in vitro and the related signaling mechanisms. Methods: TNF-α (25 ng/mL) in combination with TGF-β1 (5 ng/mL) was used to stimulate EMT of human NSCLC A549 and H460 cells. Cell proliferation was analyzed using a sulforhodamine B assay. A wound-healing assay and a transwell assay were performed to examine cell motility. Western blotting was used to detect the expression levels of relevant proteins. siRNAs were used to knock down the gene expression of c-FLIP and N-cadherin. Stable overexpression of c-FLIP L (H157-FLIP L) or Lac Z (H157-Lac Z) was also performed. Results: Treatment with TNF-α+TGF-β1 significantly enhanced the migration of A549 and H460 cells, increased c-FLIP, N-cadherin (a mesenchymal marker), snail (a transcriptional modulator) and p-Smad2/3 expression, and decreased IκB levels in the cells; these changes were abrogated by co-treatment with HNK (30 μmol/L). Further studies demonstrated that expression level of c-FLIP was highly correlated with the movement and migration of NSCLC cells, and the downstream effectors of c-FLIP signaling were NF-κB signaling and N-cadherin/snail signaling, while Smad signaling might lie upstream of c-FLIP. Conclusion: HNK inhibits EMT-mediated motility and migration of human NSCLC cells in vitro by targeting c-FLIP, which can be utilized as a promising target for cancer therapy, while HNK may become a potential anti-metastasis drug or lead compound.

Acne vulgaris is one of the most prevalent skin diseases which affect almost 80% of adolescents in the world during their lifetime. Development of specific association between antibiotic and bacteria during repeated treatment develops antibiotic resistance. Medicinal and aromatic plants are widely used as medicine in ancient time. Treatment of acne has been considered as a major research area in pharmaceutical and personal cosmetic care industries. The aim of the present work was to evaluate the phytochemical composition of Ocimum gratissimum, green synthesis of silver nanoparticle and to develop herbal topical gel formulation to treat acne. Ocimum gratissimum is selected based on its antibacterial activity. Phytochemical analysis revealed phytoconstituents such as alkaloids, flavonoids, tannins and saponins are present in the extract. Silver nanaoparticle was synthesized using 1 mM aqueous silver nitrate solution from the extracts of Ocimum gratissimum and formation of silver nanoparticle was confirmed by UV spectroscopy. The synthesized silver nanoparticles were stable, spherical shape with average particle size of 207.6 nm and the polydispersity index was found to be 0.256. Synthesized silver nanoparticles was incorporated into gel base and evaluated for its physical properties such as pH, viscosity, spreadability and antibacterial activity against Propionibacterium acne, Staphylococcus aureus and Escherichia coli. The results obtained in the developed formulation showed no lumps, had uniform color dispersion and were free from any fiber and particle. It was also observed to have easy washablity, good spreadability, pH was found to be 6.72 and 6.80 similar to pH of the skin. The antibacterial study of the developed formulation showed inhibitory activity against Propionibacterium acne, Staphylococcus aureus and Escherichia coli. Synthesized silver nanoparticle of Ocimum gratissimum showed higher activity than the extract. Hence, silver nanoparticle of Ocimum gratissiumum in aqueous gel-base can be used as an appropriate formulation for treatment of acne vulgaris.

Raktažodžiai: tradicinės papildomos medicinos priemonės, vaistiniai augalai, paprastieji spuogai. SantraukaVaistingieji augalai yra biologiškai aktyvių junginių šaltinis. Jie plačiai taikomi lėtinių ir ūmių ligų terapijai. Viena iš galimų terapijos sričių yra dermatologija. Dermatologijos srityje augalų sintetinami biologiškai aktyvūs junginiai naudojami kaip sisteminio ir vietinio poveikio priemonės. Tokia jų pritaikymo galimybė puikiai tinka kovojant su paprastųjų spuogų (aknės) atsiradimu. Tyrimo tikslas -įvertinti vaistinių augalų naudojimą tarp paprastaisiais spuogais sergančių pacientų. Tyrimas atliktas Vilniaus ir kitų Lietuvos miestų visuomenės vaistinėse. Tyrimas apėmė 500 respondentų, atsakomumas 80,6 proc. Tiriamieji buvo suskirstyti į dvi grupes: naudojančius vaistinguosius augalus (N=246) ir nenaudojančius vaistingųjų augalų (N=157). Gauti duomenys palyginti tarpusavyje nustatant statistiškai reikšmingą skirtumą tarp respondentų bei z kriterijų. Tyrimo rezultatai -visuomenės vaistinės pacientai aktyviai naudoja vaistinguosius augalus kaip papildomą tradicinę medicinos priemonę aknės gydymui ir prevencijai (65 %). Statistiškai reikšmingo skirtumo tarp vaistinguosius augalus naudojančių ir nenaudojančių respondentų tyrime nerasta. Vaistinguosius augalus dažniau renkasi respondentai, susiduriantys su paprastaisiais spuogais ilgiau negu 10 metų (p<0,05). Išvados: vaistingieji augalai yra potencialiai tinkamas terapijos būdas pacientams, kuriuos kamuoja lėtinė paprastųjų spuogų būklė. Vaistingieji augalai gali būti naudojami kaip papildomas terapijos įrankis esant ūmiai paprastųjų spuogų būklei. ĮvadasPaprastieji spuogai (Acne vulgaris) -lėtinė uždegiminė liga, kuri, remiantis atliktais tyrimais, yra trečia dažniausiai pasitaikanti dermatologinė liga 12-25 metų amžiaus asmenims [1] bei aštunta tarp visų dažniausiai paplitusių lėtinių ligų [2]. Paprastieji spuogai išsivysto dėl suaktyvėjusios lytinių hormonų veiklos brendimo laikotarpiu, Propionibacterium acnes, ir Staphylococcus epidermidis opurtunistinės infekcijos, hiperkeratinizacijos bei sebocitų proliferacijos sutrikimų [3]. A. vulgaris gydymui naudojami ne tik receptiniai vaistai, pavyzdžiui, sisteminio ir vietinio veikimo antibiotikai, retinoidai, benzoilo peroksidas, azelaino rūgštis ir adapalenas, bet ir vaistingieji augalai [4,5].Aknės gydymui ir profilaktikai naudojami augalai, kurie kaupia eterinius aliejus, flavanoidus, riebiąsias rūgštis bei antioksidantus. Eteriniai aliejai pasižymi stipriu antibakteriniu poveikiu -stabdo P.acnes ir S.epidermidis augimą ir vystymąsi, todėl sumažėja liaukų uždegimas. Šiuo poveikiu pasižymi terpinen-4-olis, cineolis, 1,8-cineolis, linalolis, eugenolis, timolis ir kt. Šiuos eterinius aliejus labiausiai kaupia Myrtaceae šeimos augalai -pražangialapė mirtenė (Melaleuca alternifolia L.) ir įvairių rūšių eukaliptai (Eucalyptus spp.) [5]. Flavanoidų, ypač hesperidino, kvercitino, apigenino, priešuždegiminis poveikis pasireiškia gebėjimu inhibuoti fermentus, aktyvuojančius prostaglandinus, leukotrienus, citokinus, ...
